Conventional constitutive expression fundamentally limits protein production yield and quality, especially for complex or toxic proteins. We provide cell lines that switch from a high-growth phase to a high-expression phase using light. By adjusting illumination, we optimize production for the highest target protein yield.

We engineer optogenetic tools into cell lines, enabling precise, light-controlled gene expression that outperforms industry-standard CMV promoters. By dynamically adjusting light intensity and patterns, Prolific enables regulation of any target gene’s expression—at any time and to any desired level, delivering superior performance and control beyond conventional systems. Here, CHO cells were stably engineered for reporter protein production under control of the constitutive CMV promoter or different light-inducible promoters in the same genomic safe harbor locus. PLight-1-6 are different light-inducible promoters that differ in their maximal expression strength and dynamic range.

Gene expression control

By attaching light-sensitive proteins to transcription factors to enable light-induced gene expression, we can deliver decoupled growth and production, tunable gene expression, and metabolic optimization. This enables increased target protein productivity, quality, and consistency along with higher viable cell densities.

Receptor control

By attaching light-sensitive proteins to dimerizing growth factor receptor components, we use light to control growth factor signaling pathways. Temporal light control over receptor activation replaces the need for costly growth factors or proteins, thereby reducing the cost of production, shortening production runs, and simplifying media requirements.
